Cranky Kong - Donkey Kong series
Arewo Stein Hakase (アレヲ・シタイン博士) - From Kaeru no Tame ni Kane wa Naru, あれ has multiple meanings, but I think it was meant to be from the 荒れる form of it (referring to someone who is rough and is angry easily). あれ can also sound rude, but I read it is colloquial; not necessarily vulgar.
Kyorozou (and to an extent, Moe/Kyorosuke) - From Densetsu no Stafy series. Kyorozou is Moe's father and is stereotypically cranky, which lead to a fall out with his wife
Kaizo (his name itself is based on his hobby) - From Tomato Adventure. A Tsundere robot who hacks and creates unofficial Gimmicks
Princess Peach is sometimes portrayed as yandere (i.e. Super Princess Peach)
Waluigi is tsundere methinks
Who else? aaaaahhh ;_;

Hmm. Off the top of my head...
Falco, from Star Fox. - Maybe not necessarily grumpy as much as sarcastic.
Nia, from Xenoblade Chronicles 2. - Here grumpiness is explained by her back story. The English dub is divisive, but personally, I think her thick Welsh accent is what makes her sassiness work so well.
Mythra, also from Xenoblade Chronicles 2. - Her attitude is explained even more profoundly through her back story if you play through Torna - The Golden Country.
Marie, from Splatoon. - OK, maybe she's not that grumpy, but she comes across that way next to the cheerful Callie.
And as usual, there are any number of Fire Emblem characters that would qualify.

A little grammatical hint: 気難しい is a い-adjective, and as such there's no need to add な to make it an adjective. ○ 気難しい人・気難しいキャラクター × 気難しいな人・気難しいなキャラクター 気難しい ( kimuzukashii) basically means "difficult to please", by the way. Shinon from Fire Emblem: Path of Radiance and Radiant Dawn immediately comes to mind. Multiple NPCs in Tokyo Mirage Sessions ♯FE are this, seeing that they're artists. #1 in mind is the director Kuen Tarachino. I'm not sure if Hellen Gravely from Luigi's Mansion 3 counts towards this.

Nice posts . Also, thanks for the advice nocturnal YL. Adding Professor Oak and Agatha from Pokémon, Rereku (and based on James Bond 007) from Tomato Adventure, as well as Zinnia from Pokémon Omega Ruby/Alpha Sapphire.
