I also see many plot twists with that, and maybe a giygas esque boss since he's crazy.

Well, for SSE2, let's begin as SSE ended. Master Hand is dead and Tabuu killed him, who was then killed himself. This leaves Crazy Hand in control of the Smash World, and with him being a bit crazy, he remakes the world into a true kitbash of Nintendo Universes with no rhyme or reason, including more Nintendo-themed enemies and even a few towns which would have speaking characters who could explain the plot better than what we got from the first one. Of course, we will have Ganondorf, Bowser, Ridley, and the Pig King each trying to take over in the power vacuum, thus making most of the foes in the game from Mario, Zelda, Metroid, and Earthbound, but with Pokemon, Pikmin, and Fire Emblem also donating a few foes. However, something weird is happening also... Mr. Game & Watch and R.O.B. are missing and Shadow Bug versions of famous Nintendo enemies, INCLUDING the ROB Squads are abound, hinting at perhaps a stronger evil hidden in the wings. Ah...Wait, wait a second... I didn't intend this to be a sequel to SSE. Merely a remake ("expansion" something-or-other) of the original (same storyline), but with more new features. I do like the towns with speaking characters part, though. I've had that in mind for a while.

The only NPCs in SSE was Palutena and apparently Navi, too. It was nice to see them again, but I'd have liked to see more NPCs... like Toadsworth, the rest of Star Fox crew, and Professor Hector.

I'm sure the horde o' Waddle Dees that got Wario and Master Hand count too.
But that's exactly what I was saying. If you've got any more ideas for NPCs, bring them on.

I think he means that there were a lot of ROBs who were NPCs.

I suppose this can vary. But most Assist Trophies really need this kind of treatment (if it can work).
Some can be cutscene devices, but others can work in gameplay.
Samurai Goroh: Maybe a simple character (not so much a boss) with an enemy-type AI/build that you fight at one point as Captain Falcon.
Tingle: Eh...this is kind of a tricky one. Your call.
